扁竹根多肽的降糖活性研究

摘要:分析研究扁竹根蛋白酶解液中主要的降糖成分。扁竹根经磷酸盐缓冲液(PBS)浸泡、硫酸铵沉淀、透析冻干的方法提取得到扁竹根粗蛋白。该粗蛋白经碱性蛋白酶Alcalase2.4L酶解14 h后,灭酶,经Sephadex G-25纯化,制备出了Fra 1Fra 2Fra 3 三个组分的扁竹根多肽,对提取的3个组分扁竹根多肽进行α-葡萄糖苷酶抑制活性试验及质谱分析。Fra 2的扁竹根多肽对α-葡萄糖苷酶抑制活性最高,其半抑制浓度(IC50值)为37.3±3.2 μg/mLFra 2的质谱分析分子量范围为265700 Da,主要为短肽。本实验验证了扁竹根多肽具有作为原料开发降糖药物的潜力,为拓展扁竹根资源的综合利用,开发降血糖保健品、药物提供实验支持。

Anti-diabetic Activity Peptides from Iris japonica

AbstractTo evaluate and validate the main anti-diabetic activity peptides from Iris japonica. Iris japonica powder were soaked in phosphatic buffer solution (PBS), and precipitated by ammonium sulfate. After dialytic and lyophilized methods, we got crude protein of Iris japonica. and purificated by Sephadex G-25 through extinguishing enzyme after 14 hours enzymolysis treatment by Alcalase 2.4 L. Obtained 3 Iris japonica peptides, named Fra 1, Fra 2 and Fra 3, respectively. All these peptides were analysed by α-glucosidase inhibitory activity experiments and MALDI-TOF mass spectrometry (MS). The Fra 2 Iris japonica peptide owes the highest activity of α- glucosidase inhibitory, which of the half maximal inhibitory concentration (IC50) were 37.3 ± 3.2 μg/mL. The MS date showed that the mass of Fra 2 were ranged from 265 to 700 Da. The results revealed that Iris japonica was a potential anti-diabetic medicinal plant can be used in health foods and medicine development.
